Collin, let me give you a different POV. When someone is not paid enough to live, they use more government services. Housing, Care, Food, etc.

I can’t tell you what the equilibrium is for a minimum wage for any given state.

When I can tell you is when companies pay less than what someone needs to live a basic life, it’s often the taxpayers that subsidize the difference

Do you really want to subsidize, with your tax dollars, any employer ?

Personally, as an employer, I would rather pay a higher wage and figure out how to be more productive and profitable to balance it out.

I literally gave raises to employees a few years ago because it was embarrassing to me that taxpayers might be subsidizing people who work for me.

It’s the same reasons I kept paying everyone during COVID and I didnt take PPP money.

The last thing I want is your tax dollars, Collin, subsidizing my employees

The other thing about increasing the minimum wage, again, IMO, everyone else faces the same rules. Their costs go up in the same manner.

Be the better entrepreneur in your segment and your business will be just fine
JUST IN: Beloved restaurant Rubio's Coastal Grill is closing 48 locations in California due to inflation and a minimum wage increase.

Don't worry, folks. The economy has never been better. Biden and @mcuban said so.

The announcement came after CA Governor Gavin Newsom signed a mum wage for fast-food workers from $16-$20.

"The closings were brought about by the rising cost of doing business in California. While painful, the store closures are a necessary step in our strategic long-term plan to position Rubio's for success for years to come," they announced in a statement.

"Those business that are not doing very well, near the shutdown margin, this increase in costs when they try to pass it on their consumers... it may be the thing that pushes them over the edge into a situation where they go out of business," said UCLA economist Brian Wheaton.

Rubio's started as a walk-up taco stand in San Diego in 1983.

Weekend hack: HackerTube. I was annoyed with YouTube's recommendations so I had Claude filter down 43M comments over 20 years of HN to 6,454 videos that keep being recommended. *Significantly* better viewing experience, ~2 hours with Opus 5.5:
hackertube.org

- Excellent (state is kept in local browser session)
- Thumbnail is a representative frame from middle of the video instead of the super duper A/B test hyper-optimized user uploads
- Underlying data from the comment analysis as one zip file, point your Claude at it and ask for your own version
- HackerTube Longs ✊

A bit more on the process:
I use Claude Code (desktop app for Mac), Opus 5.5 on Medium effort as my default. I only talk to Claude and have it do everything, the only "manual work" I helped with in tandem was in the browser (reminiscent of MenuGen): 1) I had to create some kind of API key from Google Cloud UI to query YouTube for video metadata, 2) I booted up a VPS box for serving and 3) I bought the domain name. Once the box was up I gave Claude the IP and told it update, secure, harden, deploy and optimize the code, and it did the whole thing.

Many awesome projects that would have normally taken weeks/months even can now be few-shotted for breakfast. What else are we not thinking of.

Surprise 1: Anthropic acquired a one-person biotech
.
.
.
Surprise 2: that person is me
Surprise 3: I’m here to tell you all about it

Yes, this is what you think it is; after 6 yrs of increasingly loose anonymity: the Professor Oak reveal post

…and yes, it has gotten so boring around here that I’m literally doxxing myself to spice up the timeline

-------
Ok, allow me to introduce myself. My name is Cody Tranbarger. I mostly tweet, but occasionally find time to dabble in drug dev & starting/building biotechs. My past lives span the VC, crossover, startup continuum; Prof Oak has been my way of keeping a digital foot in those worlds, despite having my physical feet planted elsewhere.

The past 18 mo, ‘elsewhere’ was Mithril Medicine, a co. I founded to develop bsAbs for LAMA2-CMD & other ECM-related genetic diseases.

Mithril was born out of hands-on building with @MikeNGladstone, @JasonPaulRhodes, @LifeSciVC, & team @atlasventure. Somehow, I made enough half-decent calls along the way (even a broken clock is right twice a day, eh?) to convince them Mithril/I warranted a small seed. While I can hardly rationalize such an outrageously cavalier bet, I’m grateful for it, & I'd partner with Atlas again in a heartbeat.

In typical AV fashion, Mithril’s primordial form was virtual, decentralized, & ultra-lean. This stage tends to be short-lived, but for Mithril, it worked surprisingly well, for surprisingly long, thanks in part to the rapid maturation of the AI tools I used liberally.

Eventually, Mithril became a bit of an experiment itself - to find the upper bound of how far it could scale as a single operator + a bunch of Claude (+ a stellar bench of part-time consultants).

In parallel, I started spending time with @nc_frey, @an1lam, & @CoefficientBio team; and promptly experienced, for the 1st time, AI at the frontier. Soon thereafter, Mithril stopped looking like an experiment, & started looking like a preview of an entirely new operating model.

From that spark came “Pipeline-in-a-Person.” The idea immediately resonated when we shared it here back in April; and it was central to the shared vision that led to Anthropic’s acquisition of Mithril, & my joining the team.

Why join Anthropic? Three reasons.
(1) For Mithril, the programs continue, with more resources & infrastructure behind them, and as much Claude as I can put to work.

(2) For me, the chance to start several more programs, & find out how many one person can actually run in parallel - an almost unprecedented luxury for a curious drug hunter.

(3) For our mission, a path to institutionalize & scale this new way of working, then put it in the hands of all of biopharma, all over the world - which, candidly, I find the most energizing.

Will AI x Bio cure diseases? Unequivocally yes, imo… but not by the route that gets the most airtime. Rather than timeline acceleration & compression, I see orchestration & parallelization as today's low-hanging fruit.

Most drug hunters see a handful of go/no-go calls in a career, & spend entirely too much of the rest trading emails (my inbox can attest).

Give them the bandwidth to run several programs at once, and the time to focus on scientific judgment, and I’m willing to bet they’ll make better calls. And at scale, better calls translate to more drugs, higher hit rates, broader patient impact, & likely, a disease or 10 or 100 cured. I couldn't be more excited to build toward that future @AnthropicAI.

Finally, a heartfelt thank you to BioTwitter/X. The end of anonymity is bittersweet, but if anyone’s earned a proper introduction, you all have.

It’s difficult to overstate how much this app has done for me: a decade of education, plenty of friends, many job opportunities, several mentors, a few investors, even a co-founder (fmr). It was the generosity of this community that inspired me to start posting 6 years ago, after just as many years lurking. I hope I’ve been able to pay it forward, in my own small way.

But, to be clear, Prof Oak isn’t going anywhere. Same beat, same themes, and now, a front-row seat to drug development at the frontier. Stay tuned for dispatches from the trenches; and remember...

Gotta Cure 'Em All

🚀 Exciting work by @brian_rini and colleagues! IRIS-RCC brings us closer to reproducible, individual-sample molecular subtyping in RCC.

We are exploring a complementary approach in mRCC: vascular architecture as a biomarker of IO sensitivity, assessed on routine H&E or CD31 IHC.

Our initial data suggest that low vessel branching may enrich for IO response, while our vascular score has also shown baseline prognostic value in CheckMate 914.

The exciting opportunity: a potentially scalable, pathology-based biomarker that could be easier to integrate into routine clinical workflows.

Next step: validation in digitized H&E from large, randomized mRCC cohorts—critically, to distinguish predictive from prognostic value.

Several groups have attempted to predict the IM151 RCC clusters with flawed methods. We developed IRIS-RCC (Individual RNA-seq Intrinsic Subtyping for RCC; available at irisrcc.vindhyadatascience.com), to predict clusters using an individual-sample model.

ESMO 2026 | A cautionary tale for immunotherapy de-escalation!

MOIO Trial (LBA10) – Phase III non-inferiority trial testing 3-monthly immunotherapy vs standard dosing in responding patients with advanced/metastatic cancer.

📊 Key results (N=308):

• Median PFS: 21.9 vs 25 dard; HR 1.35)

• Risk of death: HR 2.0 (95% CI 1.1–3.9; p=0.0143) ⚠️

• PFS HR in urothelial cancer: 2.9

• Grade 3–4 immune-related AEs: 9.5% vs 10.5%

❌ Non-inferiority NOT demonstrated.

❌ No meaningful safety advantage.

⚠️ Signal of inferior survival with reduced-frequency IO.

💡 Take-home: Less frequent immunotherapy is not necessarily equally effective!

Important: These are interim results with relatively small numbers and heterogeneous tumour types.
