Now published at @CellGenomics! Substantial amounts of new data added from the preprint version (thank you Editor and reviewers!). But the thread below still works.

TL/DR: naked RNA can enter cells & cells respond. RNases in the blood prevent that.

cell.com/cell-genomics/…
📌Preprint alert!

Can naked extracellular RNA enter cells? Can it escape from endosomes into the cytosol? Can naked extracellular mRNAs be translated if not encapsulated in EVs or LNPs?

Why do we have so many RNases in the blood? 🤔

Today in @Nature we present the development and characterisation of the first-in-class catalytic inhibitor of METTL1 as a new therapeutic strategy against solid and blood cancers.

Another flagship collaboration between my lab @SCICambridge @Cambridge_Haem (@Cambridge_Uni) and @storm_tx, co-led by my brilliant colleague @OliverRausch and driven by Eliza Yankova, Alexandra Sapetschnig and Beth Thomas.

A big thank you to the long list of authors who participated in this study and provided their critical input and diverse expertise. I am also very thankful and lucky to have the continuous support of @wellcometrust, @LeukUK and @CR_UK.

What is fascinating about academia as a career is that there are certain ideas that you have been working on since you remember. The numerical optimalities of the genetic code is what drew me to biology in the first place. I ended up working on codon usage as grad student and tRNAs as a postdoc, as the language layer on top of the genetic code. And now, 25 years later, I’m still amazed by how under-explored this field remains.

So, new preprint with Vijay Ramani 🧵
Hundreds of human tRNA genes form a redundant repertoire for decoding the genetic code. But does redundancy imply functional equivalence?

A pan-cancer in vivo CRISPRi atlas from @genophoria and Vijay Ramani at @GladstoneInst reveals a structured landscape of tRNA dependencies.

Is the RNA in our blood just fragile debris, or a readable record of how the body responds to disease? 🧬🩸 In our new Opinion in @TrendsBiochem, we propose a framework for cfRNA fragmentomics.

Adying cell releases two copies of a gene as DNA, but hundreds to millions as RNA. And the best cancer signals may come not from the tumor, but from the body's response to it: platelets, immune cells, the liver, blood vessels.

These ideas also shape our work at @B4_RNA: viewing cfRNA as a systemic response suggests why it may be especially sensitive for detecting early tumors.

A single tRNA or rRNA can be cleaved at different sites, generating small RNAs with distinct functions depending on where the cuts are made. That fragmentation pattern is itself a regulatory layer beyond abundance — the "RNA fragmentome".
With @tongzhoulab, we built qMAP, a tool to read this hidden information across development, disease & aging — for both mechanistic discovery and translational applications.

Happy to share our new PNAS paper! Building on our earlier finding that blocking endolysosomal fusion increases exosome output, we now show why: without ARL8, MVEs escape lysosomal degradation and gain secretory capacity via the RAB11A–exocyst pathway.
New study from our lab at NICHD, NIH! We identify TBC1D9 and TBC1D9B as novel ARL8 effectors that suppress #exosome secretion by inactivating RAB11A and thus blocking exocyst-dependent fusion of multivesicular endosomes with the plasma membrane. pnas.org/doi/10.1073/pn…

The only piece worth reading about Messi and Argentina right now. From the start, the hate campaign felt inorganic—and this exposes why. The implications are frightening, reaching far beyond one athlete, into how narratives are manufactured and public opinion weaponized.

One of the most amazing things I’ve ever seen: a standing ovation for the full Daraxonrasib results

I feel inspired and energised, to put it mildly — we have a targeted therapy for pancreatic cancer now, and nothing is undruggable anymore
