Looking forward to presenting updated STX-001 clinical data at Society for Immunotherapy of Cancer Annual Meeting @sitcancer
#SITC26
@MelanomaAus @Sydney_Uni
Strand is bringing two presentations to #SITC26 this week, covering our programs STX-001 and STX-003.

Fresh off her late-breaking presentation at ESMO, @ProfGLongMIA of @MelanomaAus will be presenting updated Phase I data on STX-001’s durable, systemic anti-tumor activity.

As ram, STX-001 provided preliminary evidence that a therapy injected directly into a tumor may activate the immune system throughout the body. It helped lay the foundation for STX-003, our flagship clinical program that just dosed its first cohort of patients in ASCENT-1.

Trial principal investigator, @MLJohnsonMD2 of @SarahCannonDocs, will present on the design of ASCENT-1, which is evaluating STX-003 as a systemically delivered, programmable self-replicating mRNA engineered to switch on selectively inside tumors.

📅 Friday, November 6 | Phoenix Convention Center
📢 STX-001 oral: 4:55–5:05 pm MT, West Building-300 Level, 301BC
🧬 STX-003 poster: North Building-Lower Level, Exhibition Halls 4-6

Excited for this moment! I joined @StrandTx for the mission to build programmable genetic medicines for patients in need. IL-12 has enormous promise in cancer immunotherapy, but toxicity has been a major hurdle. We're excited that STX-003 could change that. #cancer #immunotherapy
The first patients have been dosed in ASCENT-1, our Phase 1 trial of STX-003, an IV-delivered mRNA therapy designed to bring the potent immune-activating protein IL-12 to tumors throughout the body.

It’s our first systemic test of programmable mRNA, and it’s built to solve a cancer treatment for decades: the drugs powerful enough to kill tumors often damage healthy tissue, too. STX-003 is designed to sidestep that problem by selectively activating where it’s needed.

We believe it’s just the start of what the platform could make possible in this era of next-generation delivery. If this works, the same technology could carry other therapeutic proteins, opening the door to a new class of programmable medicines.

Our CEO and co-founder @DrSynbio co-founder, President, and Head of R&D @tasuku_kitada take a moment to discuss this milestone and how this trial is a test of a fundamentally new approach to treating disease.

Should PSMA PET–defined MFS count as a legitimate, actionable trial endpoint?

Dr. Steve Cho @UTMDAnderson at #USPCC26 says YES.

✅ PSMA PET outperforms CT/bone scan for nodal, bone, and visceral metastases
✅ In an EMBARK-eligible cohort with negative conventional imaging, 84% M1 disease
✅ Bone scans are becoming obsolete—more sensitive molecular imaging should define progression
⚠️ Use rigorous PROMISE/PSMA-RADS criteria, expert central reads, and confirm equivocal lesions

Bottom line: A new PSMA PET–detected metastasis is real progression—not “metastasis-free” simply because CT/bone scan is negative.

🧬 Capivasertib + abiraterone offers a precision-medicine alternative to chemotherapy-based triplet therapy in PTEN-deficient metastatic APMN/S prostate cancer.

At #USPCC26, Dr. Russell Szmulewitz @UChicagoMed highlighted CAPItello-281: in 1,012 patients with PTEN-deficient de , capivasertib improved median rPFS from 25.7 to 33.2 months, with the largest apparent benefit in complete PTEN loss. OS is still immature, while toxicity—including diarrhea, hyperglycemia, rash, and anemia—was manageable but led to discontinuation in ~18%.

Bottom line: For high-volume, de novo PTEN-deficient disease, capivasertib + abiraterone targets a poor-prognosis biology at the earliest opportunity—without requiring docetaxel.
